Breast Cancer Translational Research Laboratory J.C. Heuson

Onderzoek

Research themes

- Identification of prognostic and predictive markers in breast cancer using high throughput technologies such gene expression arrays, CGH, microRNAs, deep sequencing etc.
- Evaluation of the role of the tumor microenvironment (including immune components) in breast tumor invasion/dissemination
- Molecular characterization of the minimal residual disease (circulating (CTCs) and disseminated (DTCs) breast tumor cells
- Study of the role of the host in tumor invasion/dissemination and in the development of distant metastases (colonization)
- Study of epigenetic changes associated with breast tumorigenesis and response to different therapeutics
- Studies of functional readouts of key molecular pathways involved in breast tumorigenesis and response to different treatments including targeted agents (trastuzumab, AKT, mTOR, PI3K inhibitors etc)

Current Research Activities

1) Genetic characterization of breast cancer using the next generation of sequencing tools
2) Epigenetic characterization of breast cancer and its associated stroma
3) Role of tumor microenvironnement in breast cancer development and progression
4) Molecular characterization of the minimal residual disease (circulating (CTCs) and disseminated (DTCs) tumor cells)
5) Analysis of functional signaling output associated with major genetic aberrations in breast cancer

Achievements

1) Validation and refinement of the molecular subtyping of breast cancer (Sotiriou C. et al. Proc Natl Acad Sci U S A, 2003; cited 843 times).
2) Better characterization of histological grade by using gene expression profiling in breast carcinoma - Identification of the “Genomic Grade” (Sotiriou C. et al. J Natl Cancer Inst, 2006; cited 514 times)(Loi S. et al. J Clin Oncol, 2007; cited 207 times).
3) Meta-analysis of publicly available gene-expression and clinical data from almost 3000 breast tumors
(Desmedt C. et al. Clin Cancer Res, 2008; cited 85 times) ( Wirapati et al. Breast Cancer Res, 2008; cited 122 times).
4) Anthracyclines based chemoresistance score (Paper submitted).
